The Pensacola & Louisville Railroad Company was a railroad company incorporated by an act of Florida legislature on July 16, 1868.

It was reorganized on February 4, 1872, "having become the assignee of the Alabama and Florida Railroad of Florida," the full assets of which were sold and conveyed by A. E. Maxwell on December 10 of that year.

The railroad was sold and renamed to the Pensacola Railroad Company on May 6, 1878.
